چکیده

Absorption of digitoxin «us studied in 14 fasted anesthetized dogs after administration of 50 (JLC of H-digitoxin mixed with 1 mg of unlabeled digitoxin into either the duodenum or stomach. Radioactivity was measured in portal, hepatic, and central venous plasma, thoracic duct lymph, common duct bile and urine for 5 hours after digitoxin was given. Three dogs were given a single intravenous dose of 25 fie of H-digitoxin mixed with 0.5 mg of unlabeled digitoxin, and radioactivity was measured in central venous plasma and thoracic duct lymph. Digitoxin was absorbed rapidly and uniformly from the duodenum and entered the body entirely by way of the portal vein, reaching peak concentrations in portal blood in 1 to 15 minutes. Digitoxin was absorbed erratically from the stomach. No metabolites of digitoxin were observed in selected portal venous samples subjected to thin-layer chromatography. Absorption of orally administered digitoxin into the systemic bloodstream is retarded because of highly efficient hepatic extraction of the drug and subsequent transfer into bile.

Fate of orally administered 3H-digitoxin in man with special reference to the absorption.

3H-digitoxin and polyethylene glycol (nonabsorbable marker) were given orally to five subjects provided with a gastrointestinal tube. Of the radioactivity administered, approximately 15, 30, and 70% had been absorbed when the test solution was passing the stomach, the duodenum, and the upper jejunum, respectively. The absorption was rapid, and the plasma contained detectable amounts of radioact...

An Investigation into the Role of P-Glycoprotein in the Intestinal Absorption of Repaglinide: Assessed by Everted Gut Sac and Caco-2 Cell Line

The present study aimed at exploring the potential of the P-glycoprotein (P-gp) transporters as a barrier to the repaglinide (REG) epithelial permeability. In-vitro intestinal absorption models, the everted gut sac, and Caco-2 cell line, were used to study the possible role of P-gp in intestinal transport of REG. In the everted gut sacs, apparent permeability coefficients showed cargo concentra...

Non-absorption of ingested lipophilic and amphiphilic allelochemicals by generalist grasshoppers: the role of extractive ultrafiltration by the peritrophic envelope.

The role of the peritrophic envelope in the non-absorption of three allelochemicals ingested by generalist grasshoppers was examined. This study tested the hypothesis that the association of lipophilic and amphiphilic allelochemicals with lipid aggregates (mixed micelles) reduces their permeability through the peritrophic envelope, a process similar to extractive ultrafiltration. Each of three ...

Modeling and Experimental Study of Carbon Dioxide Absorption in a Flat Sheet Membrane Contactor

comIn the present study, CO2 removal from natural gas stream has been studied using a flat sheet membrane contactor. A three dimensional mathematical model is developed to describe the process. The model considers the transport of a gas mixture containing carbon dioxide and methane through a flat sheet membrane contactor module. The model is based on the non-wetted mode of operation, in which t...
